Laboratory diagnosis of FMD using real-time RT-PCR in Egypt

چکیده

Definitive diagnosis of foot-and-mouth disease (FMD) requires the detection of virus antigen or genome in clinical material. The aim was performance of real-time RT-PCR (rRT-PCR) procedures for this purpose. Twenty nine cattle samples of vesicular epithelial and nasal swabs from four localities of Ismailia governorates were examined by ELISA, VN, RT-PCR and rRT-PCR. The results showed that 11 samples were positive by ELISA and virus isolation, 8 of serotype O and 3 for type A. Fourteen samples out of 29 were positive by RT-PCR and rRTPCR. The features that influence sample quality appear to be less important for the rRT-PCR and RT-PCR as they can detect a small fragment of FMDV genomic RNA. Real-time RT-PCR provided an extremely sensitizer and rapid procedures that contributes to improved laboratory diagnosis of FMD. Rapid and accurate diagnosis of Foot-and-mouth disease virus by Real-time PCR in field samples

During 2010-2011, Real-time PCR procedure was used to detecting FMDV RNA on 147 epithelium samples from the field. In this survey, for Real-time PCR from 3D gene segment as conserve region selected for tracking all of seven serotypes FMDV. The assay detected the viral RNA in all serotypes of FMDV. The rRT-PCR specifically detected FMD virus in sample with greater sensitivity than our convention...

Diagnosis of Foot-and-Mouth Disease Virus by Real Time Reverse Transcription Polymerase Chain Reaction Assay in Iran

Background and Aims: Accurate and rapid diagnosis is necessary for effective control and prevention of foot-and-mouth disease (FMD). In present study, was evaluated real time reverse transcription-polymerase chain reaction (rRT-PCR) assay along with diagnostic routine methods for the detection of all seven serotypes of FMD virus (FMDV), namely O, C, A, SAT1, 2, 3 and Asia 1 in biological sample...

Enhanced laboratory diagnosis of foot and mouth disease by real-time polymerase chain reaction.

The performance of an automated real-time reverse transcription polymerase chain reaction (RT-PCR) was compared to virus isolation (VI) in cell culture and antigen detection enzyme-linked immunosorbent assay (ELISA) for the laboratory diagnosis of foot and mouth disease (FMD). The World Reference Laboratory for FMD in Woking, the United Kingdom, examined a collection of 334 epithelia received f...

Loop-mediated isothermal amplification (RT-LAMP): a new approach for the detection of foot-and-mouth disease virus and its sero-types in Pakistan

Successful disease management requires a rapid and sensitive diagnosis method that can recognize early infection even before the manifestation of its clinical signs. The only available field diagnostic tests for foot-and-mouth disease (FMD) are lateral flow devices, commonly known as chromatographic strips. Low sensitivity and inability to detect FMD virus (FMDV) at the serotype level are limit...

Diagnosis of foot-and mouth disease by real time reverse transcription polymerase chain reaction under field conditions in Brazil

BACKGROUND Foot-and-mouth disease (FMD) is an economically important and highly contagious viral disease that affects cloven-hoofed domestic and wild animals. Virus isolation and enzyme-linked immunosorbent assay (ELISA) are the gold standard tests for diagnosis of FMD. As these methods are time consuming, assays based on viral nucleic acid amplification have been developed. RESULTS A previou...
